| I. Background | |
| 由 IL-12、IL-23、IL-27、IL-35 和 IL-39 形成的細胞因子白細胞介素 12 （IL-12） 家 族在先天性和適應性免疫應答的誘導和調(diào)節(jié)中起關鍵作用。IL-27 主要由抗原呈遞細胞 （APC） 產(chǎn)生，包括樹突狀細胞 （DC）、漿細胞、巨噬細胞、炎性單核細胞、小膠質(zhì)細 胞、B 細胞和內(nèi)皮細胞。IL-27 通常參與不同 CD4 T 細胞亞群的分化和激活。受腫瘤抗原 和 CD40 刺激的抗原呈遞細胞可以分泌 IL-27，IL-27 在調(diào)節(jié)針對癌癥的免疫反應方面具有 雙重功能（促炎和抗炎作用）。IL-27 對人類病毒具有抗病毒特性，包括流感、單純皰疹病 毒、人類乙型肝炎 （HBV） 和丙型肝炎 （HCV）、人類免疫缺陷病毒 （HIV） 和其他病 毒等。 IL-27 是一種由 EBi3 和 p28 亞基組成的異二聚體細胞因子。IL-27 通過由兩個亞基組 成的異二聚體受體發(fā)出信號，即 IL-27Rα（也稱為 WSX-1 或 TCCR）和 gp130 （IL6ST）。 IL-27 結合激活 Janus 激酶 （JAK） 信號轉導和轉錄激活因子（STAT1 和 STAT3）和絲裂 原活化蛋白激酶 （MAPK） 信號轉導通路。 | |
